I romanen moder forstepersonsfortaelleren Old Shatterhand apachen Winnetou, og efter nogle indledende dramatiske begivenheder opstar der et aegte venskab mellem dem. Winnetou bliver hovding for mescalero-apacherne, efter at hans far og hans soster er blevet draebt af den hvide bandit Santer, og Old Shatterhand bliver blodbror til Winnetou. Ved mange lejligheder viser de store kampevner, men ogsa medfolelse med andre mennesker. Karl Mays roman udtrykker et romantisk onske om et enklere liv i taet kontakt med naturen. Dens popularitet skyldes i vid udstraekning fremstillingens evne til at imodekomme mange europaeeres forestillinger om det vilde Vestens mere utaemmede og autentiske miljo. Romanen var sa populaer, at Nazi-Tyskland ikke forbod den pa trods af dens heroiske syn pa farvede folkeslag; i stedet blev det argument fremsat, at historierne demonstrerede, at de amerikanske oprindelige folks fald var forarsaget af mangel pa racebevidsthed.
